Synopsis: The purpose of this study is to compare the efficacy of ZD6474 and ZD1839 in patients with NSCLC after Failure of Prior Platinum-based Chemotherapy.

Study Type: Interventional, Treatment, Randomized, Double-Blind, Uncontrolled, Crossover Assignment, Efficacy Study
Minimum Age/Maximum Age: 18 Years/
Genders: Both
Protocol Entry Criteria: Inclusion Criteria: - Prior histologic or cytologic confirmation of advanced (IIIB/IV) NSCLC - Failure of first-line platinum-based chemotherapy (the prior regimen must have failed the subject because of toxicity or progression of tumor) - WHO performance status of 0 or 1 - Life expectancy of 12 weeks or longer - Measurable disease (at least 1 target lesion) according to RECIST Exclusion Criteria: - Mixed small cell and non-small cell histology - Brain metastases or spinal cord compression, unless treated at least 4 weeks before entry and stable without steroid treatment for 1 week - Any unresolved chronic toxicity greater than CTC grade 2 from previous anticancer therapy - Prior treatment with agents that inhibit EGFR or VEGF signalling - Superior vena cava (SVC) syndrome - Significant cardiac event (including symptomatic heart failure or angina) within 3 months of entry or presence of any cardiac disease that in the opinion of the Investigator increases the risk of ventricular arrhythmia - History of clinically significant arrhythmia (multifocal PVCs, bigeminy, trigeminy,ventricular tachycardia, bradycardia) that is symptomatic or requires treatment (CTC grade 3) or a symptomatic sustained ventricular tachycardia - Chronic atrial fibrillation - Previous history of QTc prolongation while taking other medication Congenital long QT syndrome - Hypertension not controlled by medical therapy (systolic blood pressure greater than 160 mmHg or diastolic blood pressure greater than 100 mmHg)
